General Steps in Producing an Electron Micrograph:

  1. Expose the sensitive photographic emulsion by means of electrons.
  2. Develop the exposed emulsion in the darkroom to generate a negative image.
  3. reat the negative with fixer for stabilization.
  4. Wash and dry the negative.
  5. Project the negative onto a photographic paper using an enlarger.
  6. Develop the photographic paper.
  7. Stop the development process and stabilize the image in photographic fixer.
  8. Wash and dry the photographic print.
Electron Microscopes Microtomes

ZEISS 109 TEMZEISS 109 TEM
The Transmission Electron Microscope (TEM) provides information about the ultrastructure, or internal structure, of thin sections of a specimen.

 

JEOL JXA-35 SEMJEOL JXA-35 SEM
In contrast, use of the Scanning Electron Microscope (SEM) provides information about the shape and surface topography of a specimen.

MicrotomeSorvall MT-6000 Thick
Sections 0.5-2.0 um in thickness. Stain with general stains - Mallory's Azure II Methylene Blue or Toluidine Blue. Used to screen tissue, and select area to be viewed at EM level.

MicrotomeLeica Ultracut E
All three microtomes are utilized in the sectioning of tissue embedded in resin. All can be used in thick and/or thin sectioning.

 

 

Microtome Sorvall MT2-B
Thin
Sections 60-100 nm in thickness. For observation at the EM level. Stain with heavy metals - Uranyl Acetate and Lead Citrate.

Darkroom
Darkroom The Darkroom Facility is open to faculty, staff and students and can be utilized in the development/printing of electron microscopy films, light microscopy films and X-ray films.
